Dr. Alessio Rosario Marte, a neuro-immuno-gastroenterologist, has been at the forefront of experimental research into the role of the microbiota in the pathogenesis of neurodegenerative diseases. His studies have demonstrated that gut dysbiosis, characterized by a significant alteration in microbial homeostasis, is directly implicated in the pathophysiology of conditions such as Alzheimer’s disease. These findings reveal that dysbiosis triggers chronic neuroinflammation, compromises the integrity of the blood-brain barrier, and facilitates the aggregation of misfolded proteins, which are critical factors in neurodegenerative progression. In this publication, the author presents a novel therapeutic paradigm that leverages three pharmacologically distinct molecules: DDL-920, GV-971, and TRIM21, each selected to intervene in different, yet interrelated, molecular mechanisms of neurodegeneration. • DDL-920 is designed to restore gamma oscillatory activity, a neural rhythmic pattern essential for higher cognitive functions, including memory encoding and attentional processes, which are often disrupted in neurodegenerative diseases. • GV-971, a marine-derived oligosaccharide, acts on the gut-brain axis by modulating microbial dysbiosis, reducing neuroinflammation, and restoring microbial homeostasis. By directly influencing gut-derived immune responses, GV-971 mitigates the inflammatory pathways contributing to neurodegenerative pathology. • TRIM21, an engineered molecule with immunomodulatory properties, facilitates the selective degradation of hyperphosphorylated tau proteins, a critical driver of neuronal loss in diseases such as Alzheimer’s, thereby limiting tau propagation and preserving neuronal integrity. This work outlines the potential of this multi-targeted therapeutic approach not only to ameliorate cognitive impairments but also to address the underlying molecular and cellular dysfunctions driving neurodegenerative diseases. This integrative strategy represents a significant step forward in developing more effective treatments for neurodegenerative disorders.
